A combination therapy consisting of high-dose intravenous ascorbic acid (Vitamin C) and pazopanib, a multi-tyrosine kinase inhibitor. This regimen is being investigated by the Academic and Community Cancer Research United (ACCRU) for the treatment of metastatic or unresectable clear cell renal cell carcinoma (ccRCC). Pazopanib acts by inhibiting vascular endothelial growth factor receptors (VEGFR), platelet-derived growth factor receptors (PDGFR), and c-Kit, thereby blocking angiogenesis and tumor cell proliferation. Intravenous ascorbic acid is hypothesized to act as a pro-oxidant at high concentrations, generating hydrogen peroxide in the tumor microenvironment to induce oxidative stress and enhance the antitumor activity of pazopanib.
