Overview

Asedebart is a humanized monoclonal antibody designed to target and neutralize adrenocorticotropic hormone (ACTH). Developed by Ascendis Pharma, it is being investigated for the treatment of ACTH-dependent disorders, including Cushing's disease and congenital adrenal hyperplasia (CAH). By binding to circulating ACTH, asedebart prevents the hormone from interacting with melanocortin 2 receptors (MC2R) in the adrenal cortex, thereby inhibiting the downstream production of cortisol and adrenal androgens. This mechanism aims to provide a targeted approach to managing hypercortisolism and its associated morbidities. Asedebart is currently in Phase II clinical development, with studies evaluating both intravenous and subcutaneous administration routes. Preliminary data indicate that the drug is generally well tolerated and effective in reducing urinary free cortisol levels.
