Overview

ASF-1096 is a topical cream formulation containing levosalbutamol (also known as R-salbutamol), which is a β2-adrenergic receptor agonist. It has been investigated primarily for inflammatory skin disorders, particularly discoid lupus erythematosus (DLE) and other cutaneous forms of lupus erythematosus. ASF-1096 works by selectively targeting the β2-adrenoceptor of dermal inflammatory cells, including keratinocytes and various infiltrating leukocytes. When applied topically, it penetrates through the stratum corneum and binds to β2-adrenoceptors, leading to immunomodulatory effects by reducing inflammatory reactions mediated by CD4+ lymphocytes and other β2-adrenoceptor presenting leukocytes. This results in a decrease of pro-inflammatory gene products such as IL-2, TNF-α, and IFN-γ. The drug has been formulated in various concentrations for clinical testing, including 0.25%, 0.5%, and 1.0% cream formulations.
