ASHA-044 is a molecular glue protein degrader developed by Asha Therapeutics. It targets the beta-catenin pathway (CTNNB1), which is frequently mutated in colorectal cancer and other tumors. By modulating beta-catenin activity, ASHA-044 aims to overcome immune silence in tumors—an environment where immune cells are largely absent—thereby promoting anti-tumor immunity and inhibiting tumor proliferation, resistance to cell death, and metastasis. The drug was primarily investigated for preclinical efficacy in colorectal cancer models but also considered for hepatocellular carcinoma[1][2]. Its development has been discontinued at the preclinical stage.
