Overview

Asiatic acid is a naturally occurring pentacyclic triterpenoid compound primarily found in the medicinal plant Centella asiatica. It exhibits a wide range of pharmacological activities, including anti-inflammatory, antioxidant, wound healing, and anticancer effects. Mechanistically, asiatic acid acts by modulating multiple intracellular signaling pathways and transcription factors involved in cell proliferation, apoptosis, autophagy, and stress responses. In cancer models specifically, it induces cell cycle arrest (via upregulation of P53 and P21), inhibits the PI3K/AKT/mTOR/P70S6K pathway, promotes p38 signaling activation while inhibiting ERK signaling, triggers mitochondrial apoptosis (involving Bcl-2 family regulation and Caspase-3 activation), induces endoplasmic reticulum stress responses with increased reactive oxygen species generation, and suppresses autophagy through mTOR pathway inhibition[2][3][6]. Asiatic acid also stimulates wound healing by increasing collagen production[4]. It is under investigation for its potential therapeutic applications in chronic diseases such as cancer but is not an approved pharmaceutical drug.
