AsiDNA + carboplatin is an investigational combination regimen comprising **AsiDNA**, a first-in-class DNA repair inhibitor, and **carboplatin**, a platinum-based chemotherapy agent. AsiDNA acts by mimicking DNA double-strand breaks and functions as a decoy agonist, activating DNA-PK and preventing the recruitment of DNA repair enzymes involved in homologous recombination and non-homologous end joining, thereby inhibiting cellular DNA damage response. When combined with carboplatin—which induces DNA damage through DNA crosslinking—the approach is designed to synergistically enhance anticancer effects and delay or overcome resistance to carboplatin, especially in aggressive or treatment-resistant solid tumors. Preclinical and clinical studies demonstrate that the combination increases efficacy without significantly exacerbating the toxicity profile of carboplatin. The regimen is mainly under evaluation for advanced solid tumors, including triple-negative breast cancer and non-small cell lung cancer, with the developer Onxeo leading clinical trials[1][2][3][5].
