ASK0912 is a novel polymyxin derivative being developed by Jiangsu Ausun Pharmaceutical in collaboration with the Institute of Medicinal Biotechnology of the Chinese Academy of Medical Sciences. It is designed to treat serious infections caused by multidrug-resistant (MDR) Gram-negative bacteria, including carbapenem-resistant *Acinetobacter baumannii*, *Pseudomonas aeruginosa*, and *Klebsiella pneumoniae*. Like other members of the polymyxin class, ASK0912 exerts its antibacterial effect by binding to the lipid A component of lipopolysaccharide (LPS) in the outer membrane of Gram-negative bacteria. This interaction displaces divalent cations, leading to increased membrane permeability, leakage of cytoplasmic contents, and ultimately cell death. ASK0912 is specifically intended for the treatment of hospital-acquired bacterial pneumonia (HABP) and ventilator-associated bacterial pneumonia (VABP), with a design aimed at improving the safety profile, particularly regarding nephrotoxicity, compared to traditional polymyxins.
