ASKB589

ASKB589 is a humanized IgG1 monoclonal antibody that targets Claudin 18.2 (CLDN18.2), a tumor-associated antigen highly expressed in certain cancers, particularly gastric and gastroesophageal junction (GEJ) cancers[1][2][3][4]. Developed as a second-generation agent with enhanced antibody-dependent cellular cytotoxicity (ADCC), ASKB589 also mediates complement-dependent cytotoxicity (CDC) by binding to CLDN18.2 on cancer cells[3][6]. It is being evaluated as monotherapy and in combination with chemotherapy agents such as capecitabine and oxaliplatin (CAPOX), as well as with the PD-1 inhibitor sintilimab, primarily for advanced or metastatic CLDN18.2-positive gastric/GEJ cancers[5][7]. Clinical trials have shown promising efficacy and safety profiles, including high objective response rates and disease control rates in these patient populations[3][5]. The drug is currently in Phase III clinical development for gastric/GEJ cancer, with additional studies exploring its use in pancreatic cancer and other solid tumors expressing CLDN18.2[4].
