ASKG315 is a novel engineered prodrug form of the human cytokine interleukin-15 (IL-15), designed for cancer immunotherapy. It consists of a mutated IL-15 fused to an Fc fragment and linked to a masking moiety via a tumor protease-cleavable linker, allowing selective activation within the tumor microenvironment. This design aims to enhance anti-tumor immune responses by stimulating natural killer (NK) cells and CD8+ T cells while minimizing systemic toxicity. Preclinical studies demonstrated that ASKG315 has improved pharmacokinetics, extended half-life, greater tumor-selective immune stimulation, and reduced peripheral immune activation compared to previous cytokine drugs. The drug is currently in phase 1 clinical trials for advanced solid tumors as both monotherapy and in combination with pembrolizumab.
