Overview

ASKG812 is a bifunctional fusion protein drug candidate developed by AskGene Pharma for cancer treatment. It consists of an anti-PD-1 antibody fused to an engineered interleukin-2 (eIL-2) moiety. The eIL-2 component is masked to bias its activity toward PD-1 positive T effector cells via cis agonism, aiming to enhance antitumor immune responses while minimizing systemic toxicity and off-target effects. This design leverages AskGene’s proprietary SmartKine platform for selective immune modulation at the disease site. Preclinical studies have shown that ASKG812 increases activation and cytotoxicity in PD-1+ T cells with reduced activity in PD-1− T and NK cells, resulting in robust antitumor efficacy without significant toxicity compared to reference molecules. The drug is currently in preclinical development for solid tumors and other neoplasms[1][2][3][6].
