Overview

ASKG915 is a first-in-class anti-PD-1 antibody and interleukin-15 (IL-15) prodrug fusion molecule developed for the treatment of various solid tumors. It is designed as a bi-specific recombinant fusion protein that combines an anti-PD-1 antibody with an IL-15 prodrug, enabling tumor targeting via the PD-1 pathway and local activation of IL-15 at the tumor site. The molecule remains largely inactive in circulation due to protease-dependent masking of IL-15 activity, which helps minimize systemic immune activation and toxicity. Upon reaching the tumor microenvironment—where specific proteases are overexpressed—the linker is cleaved, activating IL-15 locally to stimulate immune cells such as T cells and NK cells. This approach aims to enhance antitumor immunity while reducing peripheral side effects compared to unmasked cytokine therapies. ASKG915 is administered intravenously and has demonstrated favorable pharmacokinetics and safety profiles in preclinical studies[1][2][3][6][7].
