Overview

ASN-1 is a human monoclonal antibody discovered by Arsanis in collaboration with Adimab, designed to neutralize multiple virulence factors produced by *Staphylococcus aureus*. It specifically targets alpha-hemolysin (Hla) and the S-components of four bi-component leukocidins: HlgAB, HlgCB, LukED, and LukGH. These toxins are critical to the pathogenesis of *S. aureus* infections, as they damage host cell membranes and lyse immune cells such as neutrophils. ASN-1 was developed as one of two components in the drug candidate ASN100, a 1:1 combination of ASN-1 and ASN-2. The combination was primarily investigated for the prevention of *S. aureus* pneumonia in high-risk, mechanically ventilated patients. However, clinical development of the ASN100 program was discontinued in 2018 after a Phase 2 trial failed to meet its primary efficacy endpoint.
