ASN-2 is a fully human monoclonal antibody discovered by Arsanis in collaboration with Adimab. It is designed to neutralize LukAB (also known as LukGH), a potent bicomponent leukocidin secreted by Staphylococcus aureus that targets and kills human phagocytes, including neutrophils, monocytes, and macrophages. ASN-2 was developed as one of two components of ASN100, a co-formulation with another monoclonal antibody, ASN-1. While ASN-1 neutralizes alpha-hemolysin and four other leukocidins, ASN-2 specifically addresses LukAB, which is not neutralized by ASN-1. The combination was evaluated in Phase 2 clinical trials for the prevention of S. aureus pneumonia in high-risk, mechanically ventilated patients, but the program was discontinued in 2018 following a futility analysis.
