Overview

ASN-8639 is an orally bioavailable, potent, and selective small molecule inhibitor of Protein Arginine Methyltransferase 5 (PRMT5), developed by Asieris Pharmaceuticals. PRMT5 is an enzyme that catalyzes the symmetric dimethylation of arginine residues on both histone and non-histone proteins, thereby regulating essential cellular processes such as gene transcription, RNA splicing, and protein translation. Overexpression of PRMT5 is associated with the progression of various malignancies. ASN-8639 is designed to disrupt these oncogenic pathways, leading to cell cycle arrest and apoptosis in cancer cells. It is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors, including urothelial carcinoma, cholangiocarcinoma, ovarian cancer, breast cancer, and non-small cell lung cancer (NSCLC).
