ASN004

ASN004 is an investigational antibody-drug conjugate (ADC) designed for the treatment of solid tumors. It consists of a novel single-chain Fv-Fc antibody that specifically targets the 5T4 oncofetal antigen (trophoblast glycoprotein), which is highly expressed in a broad range of malignant tumors but shows limited expression in normal adult tissues. The antibody is conjugated via a non-cleavable linker to a proprietary polymer carrying multiple auristatin analog molecules, specifically Auristatin F hydroxypropylamide, using Dolaflexin drug-linker technology. The drug-to-antibody ratio (DAR) is approximately 10–12. Upon binding to 5T4-expressing tumor cells, ASN004 is internalized and releases its cytotoxic payload, leading to potent inhibition of mitosis and cell death in cancer cells while minimizing harm to healthy tissues. Preclinical studies have demonstrated complete and durable tumor regression across various solid tumor models including lung, breast, cervical, and gastric cancers[1][2][5][6][7].
