ASN100 is a combination product consisting of two fully human monoclonal antibodies, ASN-1 and ASN-2, both of the IgG1(κ) isotype. It was developed for the prevention of Staphylococcus aureus pneumonia in high-risk, mechanically ventilated patients. The mechanism of action involves neutralization of six key S. aureus cytotoxins implicated in pneumonia pathogenesis. Specifically, ASN-1 targets alpha-hemolysin (Hla) and four leukocidins (gamma-hemolysin AB [HlgAB], gamma-hemolysin CB [HlgCB], Panton-Valentine leukocidin [PVL], and leukocidin ED [LukED]), while ASN-2 neutralizes the fifth leukocidin (leukocidin GH [LukGH]). This dual-antibody approach aims to protect lung tissue by preventing toxin-mediated damage to epithelial cells and immune cell lysis[1][2][3][4][5]. Developed by Arsanis, clinical development included Phase 1 safety studies and a Phase 2 trial for pneumonia prevention; however, the Phase 2 trial was terminated after interim analysis indicated futility[8].
