ASN512 is an orally bioavailable small molecule inhibitor of the Tropomyosin receptor kinase (TRK) family, including TRKA, TRKB, and TRKC. Developed by Jiangsu Aosaikang Pharmaceutical, it is currently in Phase 1 clinical development for the treatment of advanced solid tumors, with a particular focus on salivary gland tumors. The drug is designed to target oncogenic drivers such as NTRK gene fusions, which are prevalent in specific subtypes of salivary gland cancer like secretory carcinoma. By inhibiting the constitutive kinase activity of these fusion proteins, ASN512 aims to block downstream signaling pathways that promote tumor cell proliferation and survival.
