ASO 8-3419 is an amido-bridged nucleic acid (AmNA)-modified gapmer antisense oligonucleotide designed to target and inhibit the expression of minichromosome maintenance complex component 8 (MCM8) mRNA. MCM8, along with MCM9, plays a critical role in homologous recombination-mediated repair of double-strand DNA breaks caused by replication stress—a process that is often upregulated in cancer cells. By reducing MCM8 expression, ASO 8-3419 selectively sensitizes cancer cells to platinum-based chemotherapeutics such as cisplatin and modestly suppresses the growth of several cancer cell lines dependent on MCM8 for proliferation. In preclinical studies, ASO 8-3419 demonstrated efficacy in vitro and in vivo (xenograft models), inhibiting tumor growth and enhancing chemosensitivity with minimal toxicity. This suggests its potential as a novel anti-cancer agent or chemosensitizer for platinum compounds[1][2][3][4].
