ASO-SNORD14E is an antisense oligonucleotide specifically designed to target and inhibit SNORD14E, a small nucleolar RNA implicated in oncogenesis. SNORD14E is overexpressed in endometrial carcinoma, where it promotes tumor cell proliferation, migration, invasion, and suppresses apoptosis. ASO-SNORD14E suppresses these oncogenic effects by inhibiting SNORD14E, leading to reduced expression of aberrant FOXM1 isoforms (FOXM1b and FOXM1c) and correcting the abnormal nuclear accumulation of β-catenin associated with cancer progression. Preclinical studies using patient-derived organoids and xenograft mouse models have demonstrated that ASO-SNORD14E significantly inhibits tumor growth and enhances apoptosis in endometrial carcinoma cells. The drug's mechanism involves both regulation of FOXM1 alternative splicing (by modulating recruitment of SRSF1) and a decrease in 2'-O-methylation of FOXM1 mRNA, reducing its stability and oncogenic activity.
