ASONEP (sonepcizumab) is a humanized monoclonal antibody developed by Lpath, Inc. for the treatment of advanced, refractory renal cell carcinoma and other solid tumors. It acts by binding to and neutralizing sphingosine-1-phosphate (S1P), a bioactive signaling lipid involved in tumor angiogenesis, proliferation, and metastasis. By sequestering S1P, ASONEP inhibits the activation of S1P receptors on tumor and vascular cells, thereby disrupting the tumor microenvironment. The drug was evaluated in a Phase 2a clinical trial for patients with advanced renal cell carcinoma who had failed prior VEGF or mTOR inhibitor therapies. Although the trial demonstrated a favorable safety profile and encouraging overall survival trends, it did not meet its primary endpoint of progression-free survival. ASONEP is the systemic (intravenous) formulation of sonepcizumab, distinct from the ocular formulation known as iSONEP.
