Overview

ASP0819 is a novel, orally available small molecule that acts as a selective opener (activator) of the intermediate conductance calcium-activated potassium channel protein 4 (KCa3.1, also known as KCNN4). Its mechanism of action involves reversing abnormal nerve firing in primary sensory afferent nerves by activating KCa3.1 channels, which helps restore normal peripheral pain signal transmission. This non-opioid analgesic approach is being developed primarily for the treatment of fibromyalgia and has shown efficacy in preclinical models of fibromyalgia and other pain conditions such as osteoarthritis and visceral pain. In clinical studies, including a phase 2 trial in patients with fibromyalgia, ASP0819 demonstrated some improvement in daily average pain scores compared to placebo at certain time points.
