ASP0892 is a plasmid-based DNA vaccine developed for the mitigation of severe hypersensitivity reactions due to peanut allergy. It utilizes the LAMP-Vax platform and encodes lysosomal-associated membrane protein (LAMP) fused with major peanut allergens (Ara h 1, 2, and 3). The mechanism of action involves stimulation of Th1 immune responses to promote allergen-specific immune tolerance. ASP0892 was evaluated in phase 1 clinical trials in adults and adolescents with peanut allergy; it was well tolerated but did not show clinically meaningful improvements in food challenge outcomes. Development was discontinued after phase 1 due to lack of efficacy[1][2][5][6].
