Overview

ASP1002 is a bispecific antibody developed by Astellas Pharma that targets claudin 4 (CLDN4) and CD137 (also known as 4-1BB). Its mechanism of action involves binding to CLDN4, which is overexpressed in certain solid tumors, and simultaneously engaging CD137 on T cells. This dual targeting enhances the antitumor immune response by promoting T cell activation, proliferation, and cytokine production against CLDN4-expressing tumor cells. ASP1002 is currently being evaluated in a phase 1 clinical trial for adults with locally advanced or metastatic solid tumors that express high levels of CLDN4. Tumor types under investigation include non-small cell lung cancer (NSCLC), urothelial carcinoma (UC), colorectal cancer (CRC), pancreatic adenocarcinoma (PA), ovarian cancer (OC), triple-negative breast cancer (TNBC), and other solid tumors[1][2][6].
