Overview

ASP1128 is a potent and highly selective modulator of peroxisome proliferator-activated receptor delta (PPARδ). It is an investigational small molecule drug developed for the prevention and treatment of acute kidney injury (AKI), particularly in patients at risk following coronary artery bypass graft and/or valve surgery. The mechanism of action involves activation of PPARδ, which promotes fatty acid oxidation in mitochondria, leading to improved mitochondrial function, reduced inflammation, decreased oxidative stress, and protection against ischemia-reperfusion-induced kidney damage. Preclinical studies demonstrated that ASP1128 improves renal function and reduces injury biomarkers in animal models. Clinical trials have shown it to be safe and well-tolerated with dose-dependent pharmacokinetics and target gene engagement[1][3][4][5][7].
