Overview

ASP1570 is a novel, orally administered small molecule inhibitor of diacylglycerol kinase zeta (DGKζ), developed for the treatment of advanced or metastatic solid tumors. By inhibiting DGKζ, ASP1570 prevents the phosphorylation of diacylglycerol (DAG) to phosphatidic acid, thereby enhancing DAG-mediated signaling pathways. This mechanism leads to increased activation and function of T cells and natural killer (NK) cells, overcoming multiple immunosuppressive signals such as PD-1, transforming growth factor beta, prostaglandin E2, and adenosine. Preclinical studies have shown that ASP1570 can restore T cell activity suppressed by these signals and induce tumor growth inhibition in various models. Clinical trials have demonstrated an acceptable safety profile and early signs of clinical activity in patients with advanced solid tumors[1][3][6][8].
