ASP1570 + docetaxel is an investigational combination therapy being developed by Astellas Pharma for the treatment of advanced solid tumors, with a primary focus on non-small cell lung cancer (NSCLC). ASP1570 is a small molecule inhibitor of diacylglycerol kinase zeta (DGKζ), an enzyme that metabolizes diacylglycerol (DAG) into phosphatidic acid, thereby acting as a negative regulator of T-cell receptor signaling. By inhibiting DGKζ, ASP1570 aims to enhance and rejuvenate T-cell activation within the tumor microenvironment to promote anti-tumor immune responses. Docetaxel is a taxane-based chemotherapy agent that works by binding to and stabilizing microtubules, preventing their disassembly and leading to cell cycle arrest in the G2/M phase and subsequent apoptosis. The combination is currently being evaluated in a Phase 1/2 clinical trial (NCT05083481) for patients with advanced solid tumors, including those with NSCLC who have progressed after receiving platinum-based chemotherapy or checkpoint inhibitors.
