ASP2138 + oxaliplatin + leucovorin + fluorouracil + irinotecan is an investigational drug combination under Phase 1/1b clinical evaluation for the treatment of advanced or metastatic pancreatic adenocarcinoma and potentially other gastrointestinal cancers. ASP2138 is a bispecific monoclonal antibody targeting Claudin 18.2 (CLDN18.2), a transmembrane protein highly expressed in certain gastrointestinal cancers, and CD3, a protein found on T-cells. By simultaneously binding CLDN18.2 on tumor cells and CD3 on T-cells, ASP2138 re-directs the body’s immune cells to attack and kill cancer cells expressing CLDN18.2. The combination partners—oxaliplatin, leucovorin, fluorouracil (5-FU), and irinotecan—are established cytotoxic chemotherapeutic agents collectively known as mFOLFIRINOX, frequently used as first-line therapy for pancreatic cancer. This regimen is being studied to determine safety, tolerability, pharmacokinetics, and preliminary efficacy[1][2][3][4].
