This is an investigational combination therapy composed of **ASP2138** (a bispecific monoclonal antibody targeting Claudin 18.2 and CD3), **pembrolizumab** (a PD-1 immune checkpoint inhibitor), **oxaliplatin** (a platinum-based chemotherapeutic), **leucovorin** (a folinic acid used to enhance fluorouracil's activity), and **fluorouracil** (a pyrimidine analog antimetabolite). ASP2138 is designed to recruit T-cells to CLDN18.2-expressing tumor cells, enabling targeted immune attack. Pembrolizumab boosts immune system activity against cancer by blocking the PD-1 pathway. Oxaliplatin, leucovorin, and fluorouracil comprise the mFOLFOX6 chemotherapy regimen, which interferes with DNA synthesis in rapidly dividing cells and is a standard treatment for gastric, gastroesophageal junction (GEJ), and pancreatic cancers. This multi-agent regimen is currently in early-phase clinical trials as first-line therapy for advanced or metastatic gastric, GEJ, and pancreatic adenocarcinomas expressing CLDN18.2[1][3][6].
