ASP2390 is an investigational DNA vaccine developed by Astellas Pharma for the treatment of house dust mite (HDM) allergy. The vaccine utilizes the Lysosomal-Associated Membrane Protein (LAMP) technology, which involves fusing allergen-encoding DNA sequences with the LAMP-1 gene. This design facilitates the trafficking of the expressed allergens directly to the lysosomal compartment, enhancing their presentation via MHC class II molecules to CD4+ T cells. The primary therapeutic goal is to modulate the immune system, shifting the response from a pro-allergic Th2 profile toward a Th1 or regulatory T cell (Treg) profile, thereby inducing immunological tolerance. Clinical evaluation has focused on safety, tolerability, and the induction of HDM-specific IgG4 antibodies following intradermal administration.
