ASP2408 is a next-generation anti–cytotoxic T lymphocyte antigen-4 (CTLA4)-Ig fusion protein engineered for improved CD86 binding affinity. It was developed as an immunosuppressive agent for the treatment of rheumatoid arthritis. By selectively and potently binding to CD86, a key costimulatory ligand involved in T cell activation, ASP2408 inhibits CD28-mediated T cell costimulation, thereby suppressing immune responses. Preclinical studies demonstrated that ASP2408 has significantly higher potency than existing CTLA4-Ig fusion proteins such as abatacept and belatacept, particularly due to its enhanced selectivity and affinity for CD86 over CD80. Clinical trials showed dose-dependent target engagement with favorable pharmacokinetics and tolerability profiles in both healthy volunteers and patients with rheumatoid arthritis[1][2][6].
