ASP2998 is a TROP2-targeted immunostimulatory antibody-drug conjugate (iADC) being developed by Astellas Pharma in collaboration with Sutro Biopharma. It is unique for its dual-payload design, incorporating both a topoisomerase 1 inhibitor and a STING (Stimulator of Interferon Genes) agonist. This combination is intended to provide both direct cytotoxic effects on tumor cells and localized activation of the innate immune system within the tumor microenvironment. ASP2998 is currently being evaluated in Phase 1b/2 clinical trials for the treatment of various advanced solid tumors, including urothelial carcinoma, non-small cell lung cancer, gastric cancer, gastroesophageal junction cancer, and breast cancer. The drug is the first dual-payload program from Sutro's cell-free protein synthesis platform to enter clinical trials.
