ASP3082 + carboplatin + pemetrexed is an experimental combination therapy being investigated for the treatment of advanced solid tumors harboring the KRAS G12D mutation. ASP3082 is a first-in-class, KRAS G12D-selective protein degrader that uses proteolysis-targeting chimera (PROTAC) technology to induce E3 ubiquitin ligase-mediated ubiquitination and proteasome-mediated degradation of the mutant KRAS G12D protein. This leads to inhibition of downstream oncogenic signaling pathways[2][4][8]. Carboplatin is a well-established platinum-based chemotherapeutic that causes inter- and intra-strand DNA crosslinks, resulting in apoptosis of rapidly dividing tumor cells. Pemetrexed is a multi-targeted antifolate that inhibits several folate-dependent enzymes involved in DNA and RNA synthesis, thereby blocking cell replication. The combination aims to leverage targeted protein degradation with established cytotoxic chemotherapy for enhanced anti-tumor efficacy, particularly in cancers such as pancreatic ductal adenocarcinoma, colorectal cancer, and non-small cell lung cancer (NSCLC)[4][6].
