ASP5094 is a humanized immunoglobulin G1 (IgG1) monoclonal antibody developed to target and inhibit integrin alpha-9 (α9), a cell surface receptor implicated in the pathogenesis of rheumatoid arthritis (RA). Overexpression and activation of integrin alpha-9 on fibroblast-like synoviocytes are associated with RA disease onset and exacerbation. By antagonizing this receptor, ASP5094 was designed to reduce synovial inflammation and joint damage in RA. The drug was investigated as an intravenous therapy for patients with moderate to severe RA who had an inadequate response to methotrexate. Clinical trials demonstrated that while ASP5094 achieved high receptor occupancy and had an acceptable safety profile, it did not show efficacy compared to placebo in this patient population[1][5][3].
