Overview

ASP546C (formerly XNW27011) is an investigational antibody-drug conjugate (ADC) targeting Claudin 18.2 (CLDN18.2), a tight junction protein frequently overexpressed in gastrointestinal and other solid tumors. Developed by Astellas Pharma through a licensing agreement with Evopoint Biosciences, the drug consists of a CLDN18.2-specific monoclonal antibody conjugated to a proprietary topoisomerase I inhibitor payload via a cleavable linker. Upon binding to CLDN18.2 on the surface of cancer cells, the ADC is internalized, and the payload is released to induce DNA damage and cell death. ASP546C is currently being evaluated in Phase 1/2 clinical trials for the treatment of locally advanced unresectable or metastatic gastroesophageal adenocarcinoma, pancreatic adenocarcinoma, and other CLDN18.2-expressing solid tumors.
