Overview

ASP6294 is a PEGylated Fab' fragment monoclonal antibody developed by Astellas Pharma that acts as a nerve growth factor (NGF) inhibitor. It was investigated for the treatment of bladder pain syndrome/interstitial cystitis, osteoarthritis, and chronic back pain. The drug works by neutralizing NGF, thereby inhibiting its interaction with receptors like TrkA and p75 neurotrophin receptor (p75NTR), which are involved in the transmission of pain signals. Preclinical studies suggest that ASP6294 may provide analgesic effects with a lower risk of adverse effects compared to full-length anti-NGF IgG antibodies, particularly regarding immune complex formation and placental transfer. Clinical development included phase 1 safety studies in healthy volunteers and phase 2 trials in patients with bladder pain syndrome/interstitial cystitis; however, development for this indication was discontinued after completion of these trials.
