Overview

ASP8232 is a novel, orally administered small molecule inhibitor of vascular adhesion protein-1 (VAP-1), also known as amine oxidase, copper containing 3 (AOC3). Developed by Astellas Pharma, it was investigated primarily for the treatment of diabetic kidney disease (DKD) and diabetic macular edema (DME). The drug acts by inhibiting VAP-1 activity, which is elevated in diabetes and contributes to vascular inflammation and tissue damage. Clinical studies demonstrated that ASP8232 significantly reduced albuminuria in patients with type 2 diabetes mellitus and chronic kidney disease when added to standard care with ACE inhibitors or ARBs. Despite promising results in reducing albuminuria, development was discontinued after phase 2 trials for both DKD and DME due to strategic prioritization[1][4][5][7].
