Overview

ASP8374 is a fully human monoclonal antibody that targets T cell immunoreceptor with Ig and ITIM domains (TIGIT), an immune checkpoint receptor expressed on various immune cells, particularly tumor-infiltrating lymphocytes. By binding to TIGIT, ASP8374 blocks its interaction with ligands CD112 and CD155, thereby preventing the inhibitory signaling that dampens immune responses against tumors. This mechanism is intended to enhance T cell-mediated anti-tumor activity. Developed initially by Potenza Therapeutics and later acquired by Astellas Pharma, ASP8374 was investigated as both monotherapy and in combination with other immunotherapies (such as pembrolizumab or cemiplimab) for advanced solid tumors and recurrent malignant glioma. Clinical development has been discontinued for these indications[1][2][3][5][8].
