ASP8731 is a selective small molecule inhibitor of the transcription factor BACH1 (BTB and CNC homolog 1). BACH1 represses NRF2-mediated gene transcription, which is involved in antioxidant responses. By inhibiting BACH1, ASP8731 increases the expression of genes such as HMOX1 and FTH1, reduces inflammation and vaso-occlusion, and induces fetal hemoglobin (HbF) production. These effects are particularly relevant for sickle cell disease (SCD), where increased HbF can ameliorate disease symptoms. Preclinical studies demonstrated that ASP8731 reduced microvascular stasis, inflammation markers, and increased HbF+ cells in both animal models and human erythroid cells. The drug was developed by Mitobridge (a subsidiary of Astellas Pharma) primarily for the treatment of sickle cell anemia but development was discontinued after phase 1 trials[1][4][6][8].
