Overview

Asparaginase erwinia chrysanthemi is an enzyme-based anticancer agent derived from the bacterium Erwinia chrysanthemi. It is used primarily in combination with other chemotherapy agents to treat acute lymphoblastic leukemia (ALL) and lymphoblastic lymphoma, especially in patients who have developed hypersensitivity to E. coli-derived asparaginases[2][6][8]. The drug works by catalyzing the deamidation of the amino acid L-asparagine into L-aspartic acid and ammonia, thereby depleting circulating levels of asparagine[5][8]. Leukemic cells are unable to synthesize sufficient amounts of asparagine due to low activity of the enzyme asparagine synthetase; thus, depletion leads to inhibition of protein synthesis and cell death in these malignant cells[5][6][8]. There are both native and recombinant forms available. The recombinant form was approved by the FDA for use in patients with ALL or lymphoblastic lymphoma who are allergic to E. coli-derived products[4][6].
