Aspartyl-alanyl-diketopiperazine (DA-DKP) is a cyclic dipeptide derived from the N-terminus of human serum albumin (HSA). It is the primary active component of Ampion, a drug candidate developed by Ampio Pharmaceuticals. DA-DKP exhibits immunomodulatory and anti-inflammatory effects by inhibiting T-cell activation and reducing the secretion of pro-inflammatory cytokines such as TNF-alpha and IFN-gamma. Its mechanism of action involves the modulation of signaling pathways, potentially through interaction with the CXCR4 receptor. Ampion has been investigated in Phase 3 clinical trials for the treatment of pain associated with severe osteoarthritis of the knee, administered via intra-articular injection. It has also been explored for respiratory distress in COVID-19 patients via inhalation and intravenous routes, as well as for Long COVID and hand osteoarthritis. However, Ampio Pharmaceuticals announced the cessation of all clinical development for Ampion in 2023.
