Aspergillus oryzae trypsin is a microbial-derived serine protease obtained from the filamentous fungus *Aspergillus oryzae*. It is being developed, notably by Huadong Hospital in Shanghai, as a component of enzyme replacement therapy for patients with pancreatic exocrine insufficiency (PEI). Unlike traditional porcine-derived pancreatin, this fungal trypsin is stable over a wider pH range and exhibits resistance to degradation by gastric acid, potentially offering a more efficient alternative for protein digestion in the small intestine. Its mechanism of action involves the hydrolysis of peptide bonds within dietary proteins, specifically targeting the carboxyl side of basic amino acids such as lysine and arginine. This enzymatic activity facilitates the breakdown of proteins into absorbable peptides and amino acids, compensating for the deficiency of endogenous pancreatic proteases.
