The PolyIran PolyPill is a fixed-dose combination (FDC) medication developed by the Tehran University of Medical Sciences for the primary and secondary prevention of cardiovascular diseases. Evaluated in the large-scale PolyIran pragmatic cluster-randomized trial, this formulation typically contains four active ingredients: aspirin (81 mg), atorvastatin (20 mg), hydrochlorothiazide (12.5 mg), and either enalapril (5 mg) or valsartan (40 mg). The drug targets multiple cardiovascular risk factors simultaneously, including platelet aggregation, cholesterol synthesis, and blood pressure regulation. The PolyIran study demonstrated that the use of this PolyPill significantly reduced the risk of major cardiovascular events in Iranian adults aged 50–79 years, highlighting its potential as an effective strategy for cardiovascular disease prevention in low-income and middle-income countries.
