Overview

Aspirin + minocycline is a combination of two small molecule drugs—aspirin, an anti-inflammatory and antiplatelet agent, and minocycline, a tetracycline-class antibiotic with additional neuroprotective and anti-inflammatory properties. This combination has been investigated in preclinical models for its potential to enhance cerebroprotection by targeting matrix metalloproteinase (MMP-2 and MMP-9) activity in cerebral ischemia. It has also been studied as adjunctive therapy in bipolar depression due to their combined anti-inflammatory effects. Aspirin acts primarily through irreversible inhibition of cyclooxygenase enzymes (COX-1/COX-2), reducing prostaglandin synthesis; minocycline inhibits bacterial protein synthesis by binding the 30S ribosomal subunit but also exhibits neuroprotective actions via inhibition of microglial activation and MMPs[2][6][7].
