Aspirin eugenol ester is a novel pharmaceutical compound synthesized via the esterification of aspirin (acetylsalicylic acid) and eugenol, based on the prodrug principle. It is a crystalline, odorless solid designed to combine the anti-inflammatory, antipyretic, analgesic, antioxidant, and antithrombotic effects of its components, while reducing their gastrointestinal irritation and oxidation-related instability. Once absorbed, aspirin eugenol ester is metabolized into salicylic acid and eugenol, which act synergistically. Mechanistically, aspirin eugenol ester affects arachidonic acid metabolism pathways, reducing concentrations of key pro-inflammatory and pro-thrombotic proteins such as cyclooxygenase-1 (COX-1), cyclooxygenase-2 (COX-2), arachidonate 5-lipoxygenase (ALOX5), and coagulation factor II (FII). Additional pharmacological actions include modulation of nitric oxide synthase (NOS) and nuclear factor-erythroid 2-related factor 2 (Nrf2), providing vascular endothelial protective, anti-oxidative, and anti-apoptotic effects. Investigations in animal models have demonstrated efficacy for the treatment and prevention of inflammation, thrombosis, hyperlipidemia, atherosclerosis, pain, fever, and oxidative vascular injury[1][2][3][4][5][6][7][8][9].
