AST2662 is a potent and selective small-molecule inhibitor of the RET (Rearranged during Transfection) receptor tyrosine kinase, developed by Elis Pharmaceutical Technology (also known as Allist Pharmaceuticals). It is designed to target oncogenic RET alterations, including fusions and mutations, which are key drivers in various malignancies such as non-small cell lung cancer (NSCLC) and medullary thyroid cancer. AST2662 is currently in Phase 2 development for several indications, including first-line and second-line treatment for RET fusion-positive solid tumors, as well as RET-mutant thyroid cancer. Uniquely, the pipeline also includes development for RET-independent solid tumors, which may explore the drug's utility in overcoming bypass resistance mechanisms in other oncogene-driven cancers or its activity in broader oncogenic contexts.
