Astragalus membranaceus saponins are a group of triterpenoid glycosides (saponins) extracted from the root of Astragalus membranaceus, a plant widely used in traditional Chinese medicine. The main active constituents include various astragalosides (notably astragaloside IV), which have been shown to possess immunomodulatory, antioxidant, anti-inflammatory, and anticancer properties[2][3][4][8]. Mechanistically, these saponins modulate multiple cellular pathways including inhibition of tumor cell proliferation and migration; induction of apoptosis via caspase 3 activation; cell cycle arrest at S and G2/M phases; suppression of cyclin-dependent kinase activity; regulation of the NOX4/Nrf2 signaling pathway (notably in neuroprotection/ferroptosis); modulation of Wnt/beta-catenin and Notch1/hes1 signaling pathways for osteogenic and cardioprotective effects; downregulation of VEGF for anti-angiogenesis; enhancement of immune responses through activation or regulation of toll-like receptor 4-related MAPK activities[4][5][6][8]. These compounds are under investigation for their potential use as adjuvant therapies in cancer treatment (especially gastrointestinal cancers), neurodegenerative diseases such as Alzheimer’s disease, diabetes mellitus type 2, inflammatory conditions like colitis, and as general immunostimulants[1][5][6].
