Asunercept is a fully human recombinant fusion protein consisting of the extracellular domain of the CD95 receptor (also known as Fas or APO-1) fused to the Fc portion of a human IgG1 antibody. It acts by selectively binding and inhibiting the CD95 ligand (CD95L/FasL), thereby blocking its interaction with the CD95 receptor. This blockade prevents apoptosis of activated T cells in tumors and inhibits tumor immune escape mechanisms. In oncology, this mechanism is intended to enhance anti-tumor immunity by preserving immune cell function within the tumor microenvironment. In myelodysplastic syndromes (MDS), inhibition of CD95L signaling rescues erythroid progenitors from apoptosis in bone marrow. Asunercept has also been investigated for viral infections such as COVID-19 due to its potential to reduce inflammatory cell death and lymphopenia associated with acute respiratory distress syndrome (ARDS). The drug has demonstrated good tolerability in clinical trials and has orphan drug status for glioblastoma multiforme and MDS in both Europe and the US[2][3][4][5].
