Overview

AT-002 is an investigational peptide mimetic developed by Arctic Therapeutics that modulates the Janus kinase-signal transducer and activator of transcription (JAK-STAT) pathway. The drug is designed to mimic the kinase inhibitory region (KIR) domain of the suppressor of cytokine signaling 1 (SOCS1) protein, which serves as a natural negative regulator of cytokine signaling. By mimicking this domain, AT-002 binds to and inhibits Janus kinases, thereby suppressing downstream cytokine responses and inflammatory signaling. It is currently in Phase I development for autoimmune uveitis and juvenile dermatomyositis, with additional preclinical exploration in systemic lupus erythematosus, psoriasis, and cytokine storm syndromes, including those associated with COVID-19 and CAR-T cell therapy. The drug has also shown efficacy in veterinary studies for equine uveitis.
