AT-0174 is a **novel, orally administered, dual inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO2)**, enzymes that mediate tryptophan catabolism in the kynurenine pathway. By simultaneously inhibiting IDO1 and TDO2, AT-0174 reduces kynurenine production and counteracts tumor-associated immunosuppression, specifically decreasing regulatory T cells (Tregs) and increasing cytotoxic CD8+ T cells and natural killer cell infiltration in the tumor microenvironment. This metabolic reprogramming enhances anti-tumor immunity and exhibits *synergy* with chemotherapy (temozolomide) and immune checkpoint inhibitors (e.g., anti-PD1), significantly improving survival in preclinical models of glioblastoma and treatment-resistant non-small cell lung cancer. AT-0174 is being developed for a broad range of solid tumors, including glioblastoma, NSCLC, triple-negative breast cancer, and ovarian cancer, among others[1][2][5][6].
