AT-07 is a pan-amyloid removal (PAR) therapeutic candidate being developed by Attralus Therapeutics for the treatment of neurodegenerative disorders, including Alzheimer's disease, Parkinson's disease, and Lewy body disease. It consists of a peptibody (AT-04)—a fusion of a proprietary PAR-peptide with a human IgG1 Fc component—conjugated to Ossianix's TXP1 brain shuttle, a VNAR antibody-based blood-brain barrier (BBB) shuttle that utilizes the transferrin receptor to enhance central nervous system penetration. AT-07 is designed to bind with high affinity to multiple pathogenic protein aggregates, including amyloid-beta (Aβ), tau, and alpha-synuclein (α-synuclein) fibrils, facilitating their clearance through phagocytosis. Preclinical data has demonstrated that the base peptibody AT-04 exhibits sub-nanomolar binding to these targets, while the AT-07 formulation shows a 10-fold increase in brain penetration compared to the unconjugated biologic.
